That giant 58 Remington is the largest, but I don't know if it would qualify as the most powerful.

 

There are at least two chambered in 50 BMG - a single shot and I believe a revolver. There's a revolver chambered in 600 nitro Express. I think one of them would be real close to the top.

My guess is the Noreen ULR 50BMG pistol.  A single shot bolt action with 16" barrel.  The cartridge does give up half it's energy with the short barrel.
